Research Interest

I am interested in understanding the role of sarcoplasmic reticulum (SR) calcium in cardiac physiology and pathology, particularly in SR calcium driven cardiac arrhythmias such as Catecholaminergic Polymorphic Ventricular Tachycardia (CPVT). My studies focus on mechanisms involved in either triggering or facilitating these adrenergic arrhythmias, using mouse models of CPVT.

One of the current research directions is to investigate if aerobic exercise training can offer a novel therapeutic option to improve SR calcium cycling and thereby impact triggered arrhythmias in CPVT hearts. Calcium is removed from the cytosol after contraction into the SR compartment by the Sarcoplasmic Reticulum calcium ATPase (SERCA2a). This calcium is buffered within the SR by Calsequestrin (CASQ2), the major SR calcium buffering protein. During the next cycle, calcium is released via the Ryanodine receptor (RyR2) to initiate contraction. Since SR calcium plays a key role in defining myocyte contraction, defects in calcium release lead to abnormal membrane depolarizations, which could give rise to premature ventricular beats. CPVT is characterized by defects either in the RyR2 or CASQ2, both leading to “leaky” RyR2 channels and fatal arrhythmias. Aerobic exercise is advocated as a therapy along with pharmaceutical interventions for patients with heart disease since it can increase exercise capacity, cardiac function and increase survival rates. Therefore, we are investigating if aerobic exercise can potentially help CPVT patients despite the SR calcium abnormalities. Using a mouse model of CPVT, we are studying how exercise impacts SR calcium cycling in the context of a defective release complex and in turn, arrhythmogenesis.
